  3. Vancomycin-Resistant Enterococcus faecium Infections: Nezkil tablet is used for the treatment of infections caused by vancomycin-resistant Enterococcus faecium (VRE). It can be used for complicated skin and soft tissue infections and bloodstream infections caused by VRE.
  4. Methicillin-Resistant Staphylococcus aureus (MRSA) Infections: Nezkil tablet is effective against MRSA infections, including skin and soft tissue infections, pneumonia, and complicated infections where other treatment options are limited.

Off-label Uses of Nezkil Tablet

  1. Gram-Positive Infections: Nezkil tablets may be used off-label to treat other gram-positive bacterial infections, such as infections caused by Streptococcus species, including Streptococcus pneumoniae, and other susceptible gram-positive bacteria.
  2. Bone and Joint Infections: Nezkil tablets may be considered for the treatment of bone and joint infections caused by susceptible gram-positive bacteria, including MRSA.

Who Should Not Use Nezkil?

Nezkil has certain contraindications, which are situations in which the use of Nezkil is not recommended due to potential risks.

Hypersensitivity: Nezkil is contraindicated in individuals with known hypersensitivity or allergy to Nezkil or any of its components. Serious allergic reactions, including anaphylaxis, have been reported with Nezkil use.

Use of Monoamine Oxidase Inhibitors (MAOIs): Nezkil is contraindicated in individuals taking or within two weeks of taking any form of MAOIs, including certain antidepressants and certain antibiotics. Concurrent use of Nezkil and MAOIs can result in serious and potentially life-threatening serotonin syndrome.

Serotonin Syndrome: Nezkil is contraindicated in individuals receiving or within two weeks of receiving serotonergic drugs, including selective serotonin reuptake inhibitors (SSRIs), serotonin-norepinephrine reuptake inhibitors (SNRIs), tricyclic antidepressants, and other serotonergic drugs. Concurrent use can increase the risk of serotonin syndrome, a potentially life-threatening condition characterized by altered mental status, autonomic instability, neuromuscular abnormalities, and gastrointestinal symptoms.

Uncontrolled Hypertension: Nezkil should be used with caution in individuals with uncontrolled hypertension or severe hypertension, as it can potentially increase blood pressure.

How Nezkil Works?

Binding to the 50S Ribosomal Subunit: Nezkil binds to the 23S ribosomal RNA of the 50S subunit of the bacterial ribosome, specifically at a site called the peptidyl transferase center. This prevents the formation of the initiation complex and interferes with the assembly of the ribosome.

Inhibition of Protein Synthesis: By binding to the ribosome, Nezkil blocks the formation of the functional ribosome complex needed for protein synthesis. This inhibits the elongation of the growing peptide chain and prevents the formation of new bacterial proteins.
